Item Incremento germinativo da semente do café (Coffea arabica L.): aplicação de um campo magnético uniforme com monitoramento pelo biospeckle laser(Universidade Federal de Lavras, 2018-10-22) Azevedo, Roberto Luiz de; Braga Júnior, Roberto Alves; Guimarães, Renato Mendes; Toorop, Peter EdwinUm dos grandes empecilhos na formação e produção de mudas de café (Coffea arábica L), reside no fato de as sementes apresentarem uma germinação lenta e não uniforme, o que impacta diretamente na qualidade dessas mudas e no tempo hábil de seu plantio no campo. Pesquisas sobre tratamentos pré-germinativos das sementes de café (Coffea arabica L.), têm sido realizadas no sentido de minimizar o tempo de germinação ou melhorar a uniformidade das mudas e, consequentemente, repercutindo na eficiência das lavouras. Dessa forma, o objetivo desta pesquisa foi avaliar os efeitos sobre a germinação das sementes de café, pela aplicação de um campo magnético constante como um condicionamento pré-germinativo. As análises pós-tratamento magnético foram por meio do Biospeckle Laser (BSL) consorciadamente aos testes tradicionais de viabilidade de sementes. Foram utilizadas 3000 sementes submetidas a campos magnéticos de intensidade constante e valores de 10 mT e 28 mT por um mesmo intervalo de tempo de 6 dias, durante seu processo de germinação em germinadores. Iluminou-se com luz laser a região do embrião e as imagens obtidas pelo Biospeckle Laser (BSL) foram processadas e os resultados dos níveis de atividade dessa região foram, posteriormente, comparados com os dados obtidos pela análise fisiológica tradicional de sementes. Além dos resultados de atividade do BSL, foram obtidos outros resultados das análises de sementes, durante o processo de germinação, tais como: IVE, isoenzimático (SOD, CAT, EST, MDH e endobetamananase), integridade de membranas, germinação, IVG e protrusão. Concluiu-se que o pré-tratamento magnético com ambas as intensidades de campo, durante os primeiros 6 (seis) dias de germinação, propiciou uma melhora na permeabilidade das membranas celulares, uma ativação precose do sistema antirradicular, além de promover uma germinação mais rápida e uniforme das sementes.Item Magnetic field in coffee seed germination(Editora UFLA, 2020-07-13) Braga Júnior, Roberto Alves; Azevedo, Roberto Luiz de; Guimarães, Renato Mendes; Reis, Leandro VilelaThe effective production of coffee seedlings faces many challenges, including seed germination. Therefore, a reduced seed germination period can be one of the most relevant contributions to enhance the testing and production of robust seedlings. The objective of this research was to evaluate the effects of a constant magnetic field on the germination of coffee seeds (Coffea arabica L.). The analyses were performed using a biospeckle laser (BSL) in conjunction with traditional seed viability tests. The coffee seeds were subjected to magnetic fields of constant intensity at values of 10 mT and 28 mT for a time interval of 6 days during their germination process. The embryo region was illuminated, and the images obtained by the BSL were processed. The activity levels of this region were compared with the data obtained using traditional physiological seed analysis. In addition to the results of BSL activity, the results of the seed analysis, such as isoenzymatic catalase (CAT), esterase (EST), superoxide dismutase (SOD), malate dehydrogenase (MDH) and endo-β-mananase, membrane integrity, germination, germination speed index (GSI), emergence speed index (ESI), and radicular protrusion levels, were obtained during the germination process.
