Resumo
Os experimentos foram realizados no laboratório de Cultura de Tecidos da ESAL - Lavras, MG. O material vegetal utilizado foi café Coffea arabica L. cv. Catuaí Vermelho CH 2077-2-5-44, cujas plântulas já estavam estabelecidas "in vitro" com cerca de 6 meses de idade. Para os trabalhos de micropropagação foram usados nós com folhas excisadas, enquanto que para a indução dos calos foram usadas folhas cortadas à metade. O meio de cultura básico foi o "MS" e o agente gelificante foi ágar (7g/l) com pH 5,9 antes da autoclavagem. No primeiro estudo, as concentrações dos componentes de "MS" foram 25%, 50%, 75% e 100% e, níveis de BAP (0, 3, 6 e 9mg/1): no segundo estudo, o tamanho do explante variou em 2, 4 ou 6 gemas e
O, 3, 6 e 9mg/1 de BAP; e, no terceiro, os sais de "MS" foram usados na metade de sua concentração e os outros componentes integralmente usados, acrescido de 2g/1 de caseína hidrogenada variando os níveis de cinetina (O; 0,O1; 0,1; 1,0 e 10,0mg/1) e 2,4-D (0,1; 1,O e 10mg/1). Para a micropropagação foram avaliados número total de brotos e folhas, aos 30, 60, 90 e 120 dias e, número de brotos maiores que 1cm, cor e peso da materia seca aos 120 dias, enquanto para a indução de calos foram avaliados por sistema de notas para tamanho e fenótipo dos calos aos 25, 50, 100 e 120 dias. O delineamento experimental foi DIC e quando havia avaliações em diferentes tempos, o Parcela subdividida no tempo. Para o experimento testando diferentes níveis de componentes de "MS" e BAP, a produção de brotos é maior no uso do "MS" usado integralmente na presença de 6,29mg/1 de BAP quando avaliados aos 90 dias e, 9mg/1 de BAP, quando as análises foram
feitas aos 120 dias. Para a produção de folhas, 6,17mg/1 de BAP quando avaliados aos 90 dias e 9,0mg/1, aos 120 dias, com "MS" integral. Na variável peso da matéria seca, os componentes de "MS" usados em 75% das suas concentrações e 9,0mg/1 de BAP apresentaram o melhor resultado. Para numero de gemas e níveis de BAP, a produção de brotos e folhas é melhor com 5,05mg/1 de BAP e explantes com 2 pares de gemas, enquanto que para os outros parâmetros avaliados, os melhores resultados foram conseguidos com 9,0mg/1 de BAP e explantes com 3 pares de gemas. Na induçao de calos para o parâmetro tamanho como para fenótipo, o nível de 2.4-D que produz melhores resultados é de 5,4mg/1 enquanto a cinetina não mostrou efeito na produção de calos em nenhuma concentração estudada. O melhor tempo de crescimento dos calos,
para todos os níveis de 2,4-D, esteve entre 88 e 89 dias.
The studies were realized at the tissue culture laboratory at ESAL, Lavras, MG. The vegetable material used was coffee - Coffea arabica L. cv. Catuaíi Vermelho CH 2077-2-5-44, wich seedlings were already stablished "in vitro" aged 6 months. For the micropropagation studies it was used nos with excised leaves and for the callus induction, were used leaves cutted on half. The basic culture medium was the "MS" and the gelifing agent was the ágar (7g/l) with pH 5,9 before autoclaving. An the first study, the concentrations of the components of "MS" was 25%, 50%, 75% e 100% and, BAP levels (0, 3, 6 and 9 mg/l); on the second study, the size of the explant had among 2,4 or 6 buds and 0, 3, 6 and 9mg/l of BAP; and on the third, the salts of "MS" was used a half of concentration and the other components were used integrally added 2g/l of hydrolised caseine changing the levels of kinetin (0; 0,01; 0,1; 1,0 and 10,0 mg/l) and 2,4-D (0,1; 1,0 and 10,0 mg/l). For the micropropagation, it was assessed the total number of shoots and leaves, on 30, 60, 90 and 120 days, and number of shoots bigger than 1cm, color and weight of dryed material on 120 days; while for the callus induction it were for the system of notes for size and fenotipe of the callus at 25, 50, 100 and 120 days. The experimental delineament was DIC, and when there was avaliation on different times, split plot in time. For the experiment testing different leavel of "MS" and BAP components the production of shoots is better using "MS" integrally, on presence of 6,29 mg/l of BAP when avaliated on 90 days and, 9,0 mg/l of BAP when the analysis were done on 120 days, with integral "MS". On the variable dryed material weight, the "MS" components used in 75% of its concentrations and 9,0 mg/l of BAP show the best results.
