Adição de cafeína ao sêmen suíno resfriado ou descongelado

Resumo
Avaliaram-se os efeitos da adição de cafeína ao sêmen suíno após o resfriamento e o descongelamento. No experimento 1, 12 ejaculados provenientes de quatro animais foram processados em doses de 100 mL com três bilhões de espermatozoides. Foi utilizado um delineamento em blocos casualizados (ejaculado) em esquema fatorial 5x3 (cinco doses de cafeína 0, 2, 4, 6 e 8 mM/dose inseminante e três tempos de armazenamento a 15 oC – 0, 24 e 48 horas) com parcela subdividida no tempo (tempos de avaliação), com 12 repetições de um ejaculado. A cafeína foi adicionada logo após a incubação de amostras de sêmen e as avaliações foram feitas 10 e 120 minutos depois. No experimento 2, 12 ejaculados provenientes de quatro animais foram coletados e armazenados em palhetas de sêmen de 0,5 mL contendo 500 milhões de espermatozoides. Foi utilizado um delineamento em blocos casualizados (ejaculado) com cinco tempos de avaliação após a adição de cafeína (0, 15, 30, 45 e 60 minutos), com 12 repetições de um ejaculado. Não houve efeito (P>0,05) da cafeína na motilidade, na taxa de degradação da motilidade espermática e no total de alterações morfológicas, em ambos os experimentos. Não houve efeito (P>0,05) da concentração de dialdeído malônico espermático para o sêmen resfriado. Houve interação (P<0,05) entre cafeína e tempo de armazenamento no vigor e na viabilidade espermática, tendo o vigor melhorado (P<0,05), com a adição de cafeína na concentração de 8 mM/dose, apenas no sêmen resfriado armazenado por 48 horas, em que, em tempos inferiores de armazenamento, a cafeína reduziu (P<0,05) de forma linear a viabilidade espermática. Não houve efeito (P>0,05) da cafeína na viabilidade espermática para o sêmen descongelado, mas a taxa de resistência osmótica diminuiu com o aumento da dose de cafeína utilizada (P<0,05). Não houve diferenças para a taxa de resistência osmótica e o tempo de armazenamento do sêmen (P>0,05). Conclui-se que a adição de cafeína ao sêmen suíno resfriado na dose de 8 mM melhora o vigor às 48 horas de resfriamento, porém, afeta negativamente em tempos inferiores de armazenamento e que a adição de cafeína não melhora a qualidade do sêmen suíno descongelado.
We evaluated the effects of adding caffeine to boar semen after cooling and thawing. In experiment 1, 12 ejaculates from four animals were processed in doses of 100 mL with three billion sperm. We used a randomized block design (ejaculate) in factorial 5x3 (five doses of caffeine 0, 2, 4, 6 and 8 mM/insemination dose and three days of storage at 15 °C - 0, 24 and 48 hours) with plot subdivided in time (time of review) with 12 repetitions of one ejaculate. Caffeine was added after incubation of semen samples and assessments were made at 10 and 120 minutes after. In experiment 2, 12 ejaculates from four animals were collected and stored in straws of semen containing 500 mL of 0.5 million sperm. We used a randomized block design (ejaculate) with five evaluation times after addition of caffeine (0, 15, 30, 45 and 60 minutes) with 12 repetitions of one ejaculate. There was no effect (P>0.05) of caffeine on motility, the rate of degradation of total sperm motility and morphological changes in both experiments. There was no effect (P>0.05) for the concentration of malonic dialdehyde sperm for cooled semen. There was an interaction (P<0.05) between caffeine and storage time on sperm viability and vigor, and the vigor has improved (P<0.05) with the addition of caffeine in 8 mM concentration/dose only in cooled semen stored for 48 hours, where at inferior storage times, caffeine reduced (P<0.05) linearly sperm viability. There was no effect (P>0.05) of caffeine on sperm viability for thawed semen, but the rate of osmotic resistance decreased with increasing dose of caffeine used (P<0.05). There were no differences in the rate of osmotic resistance and time of sperm storage (P>0.05). We conclude that the addition of caffeine to swine semen cooled at the rate of 8 mM improves the force at 48 hours of cooling, but negatively affects in lower storage times and that the addition of caffeine does not improve the quality of boar semen thawed.
