CARACTERIZAÇÃO DA REGIÃO PROMOTORA DO GENE DREB2A DE GENÓTIPOS DE COFFEA CANEPHORA CONTRASTANTES PARA A TOLERÂNCIA À SECA

Abstract

Apesar de alguns estudos em fisiologia vegetal resultarem em uma melhor compreensão dos mecanismos envolvidos na tolerância à seca em cafeeiro, ainda é escasso o conhecimento acerca das alterações metabólicas e moleculares envolvidos na resposta do cafeeiro às condições de estresse hídrico. Neste sentido, resultados prévios obtidos, analisando-se a expressão relativa do gene DREB2A dos clones 14 (tolerante a seca) e 22 (sensível a seca) de Coffea canephora var. Conilon, demonstraram expressão diferencial desse gene em folhas de plantas submetidas ou não ao estresse hídrico. Polimorfismos na região promotora do gene DREB2A foram identificados e podem indicar a participação de diferentes haplótipos no controle genético da tolerância a seca. O objetivo deste trabalho consiste na engenharia de cassetes gênicos utilizando o vetor binário pBI121 combinado com diferentes segmentos da região promotora do gene DREB2A, isolados dos clones 14 e 22 de C. canephora com o intuito de permitir uma melhor caracterização in vivo, da participação dos elementos cis e da influência dos polimorfismos observados na funcionalidade deste promotor em relação às respostas do cafeeiro ao estresse hídrico.

Although some studies in plant physiology has resulted in a better understanding of the mechanisms involved in drought tolerance in coffee, there are still lack of information about the metabolic and molecular mechanisms involved in the response of coffee to water stress conditions. Thus, previous results obtained by analyzing the relative expression of the gene DREB2A of the clones 14 (drought tolerant) and 22 (drought susceptible) of C. canephora var. Conilon showed differential expression of this gene in the leaves of plants subjected or not to water stress. Polymorphisms inside the DREB2A promoter region were identified and could indicate the involvement of different haplotypes in the genetic control of drought tolerance. The objective of this work consists of engineering gene cassettes using the binary vector pBI121 combined with different segments of the gene promoter DREB2A, isolated from the clones 14 and 22 of C. canephora in order to better characterize in vivo the involvement of cis elements and the influence of the polymorphism found in the functionality of this promoter upon the coffee response to water stress.
