Item Chemical diversity in coffee species of genebank of Instituto Agronômico do Estado de São Paulo(Crop Breeding and Applied Biotechnology, 2005-09-20) Aguiar, Adriano Tosoni da Eira; Fazuoli, Luiz Carlos; Salva, Terezinha de Jesus Garcia; Favarin, José LaércioThe objective of this work was to group different coffee species present in the genebank of the Instituto Agronômico by using some chemical variables. A total of thirty-nine plants belonging to seven species were analyzed for chemical seed components (soluble solids, lipids, trigonelline, chlorogenic acids and caffeine). The results evidenced that species could be grouped by lipids, chlorogenic acid, trigonelline and caffeine. The existence of great variation among and within species ranging from 24.12 to 30.65% for soluble solids; 8.68 to 17.49% for lipids; 0.32 to 2.15% for trigonelline; 2.91 to 6.38% for chlorogenic acid and 0.80 to 2.50% for caffeine was also observed, indicating the possibility to select plants of interest for the improvement of cultivated coffee species.Item Variação no teor de lipídios em grãos de variedades de Coffea canephora(Empresa Brasileira de Pesquisa Agropecuária - Embrapa, 2005-12) Aguiar, Adriano Tosoni da Eira; Salva, Terezinha de Jesus Garcia; Fazuoli, Luiz Carlos; Favarin, José LaércioO objetivo deste trabalho foi estudar a variação do teor de lipídios em grãos de diferentes variedades de Coffea canephora Pierre, do Banco de Germoplasma de Café, do Instituto Agronômico, Campinas, SP, Brasil. Foram analisadas sementes de 47 plantas pertencentes às variedades Robusta (3), Kouilou ou Conilon 66 (13), Apoatã (12), Guarini (8), Laurentii (7) e Bukobensis (4). A quantidade de matéria graxa foi determinada após extração com éter de petróleo. Os resultados revelaram diferença entre o teor médio de lipídios das variedades Kouilou (7,33%) e Robusta (10,91%). Foram constatadas diferenças também dentro das plantas das seis varieda- des, com amplitudes de 15,57% a 21,63% em Apoatã e Laurentii, respectivamente.Item Gene expression profile during coffee fruit development and identification of candidate markers for phenological stages(Empresa Brasileira de Pesquisa Agropecuária - Embrapa, 2012-07) Gaspari‐Pezzopane, Cristiana de; Bonturi, Nemailla; Guerreiro Filho, Oliveiro; Favarin, José Laércio; Maluf, Mirian PerezThe objective of this work was to identify genes that could be used as suitable markers for molecular recognition of phenological stages during coffee (Coffea arabica) fruit development. Four cultivars were evaluated as to their differential expression of genes associated to fruit development and maturation processes. Gene expression was characterized by both semi‐quantitative and quantitative RT‐PCR, in fruit harvested at seven different developmental stages, during three different seasons. No size polymorphisms or differential expression were observed among the cultivars for the evaluated genes; however, distinct expression profiles along fruit development were determined for each gene. Four out of the 28 evaluated genes exhibited a regular expression profile in all cultivars and harvest seasons, and, therefore, they were validated as candidate phenological markers of coffee fruit. The gene α‐galactosidase can be used as a marker of green stage, caffeine synthase as a marker of transition to green and yellowish‐green stages, and isocitrate lyase and ethylene receptor 3 as markers of late maturation.Item Diversidade química de cafeeiros na espécie Coffea canephora(Instituto Agronômico (IAC), 2005-10) Aguiar, Adriano Tosoni da Eira; Fazuoli, Luiz Carlos; Salva, Terezinha de Jesus Garcia; Favarin, José LaércioEste trabalho teve por objetivo caracterizar seis variedades de C. canephora do Banco de Germoplasma de Café do Instituto Agronômico, em Campinas. Para tanto, considerou-se a caracterização química de quarenta e sete exemplares analisando-se as variáveis sólidos solúveis, lipídios, trigonelina, ácidos clorogênicos e cafeína nas sementes. Observou-se a existência de grande variação entre e dentro dos diferentes materiais analisados, com valores extremos de 24,53% a 30,68% para sólidos solúveis; 6,61% a 12,27% para lipídios; 0,73% a 1,59% para trigonelina; 3,30% a 6,30% para ácidos clorogênicos e 1,94% a 3,29% para cafeína, indicando a possibilidade de seleção de plantas de interesse para o melhoramento dessa espécie.
