    Antioxidative responses of cell suspension cultures of two Coffea arabica varieties to low aluminum levels at pH 5.8

    The effects of aluminum (Al) on the activities of antioxidant enzymes and ferritin expression were studied in cell suspension cultures of two varieties of Coffea arabica, Mundo Novo and Icatu, in medium with pH at 5.8. The cells were incubated with 300 μM Al3+, and the Al speciation as Al3+ was 1.45% of the mole fraction. The activities of superoxide dismutase (SOD), catalase (CAT), and glutathione S-transferase (GST) were increased in Mundo Novo, whereas glutathione reductase (GR) and guaiacol peroxidase (GPOX) activities remained unchanged. SOD, GR, and GST activities were increased in Icatu, while CAT activity was not changed, and GPOX activity decreased. The expression of two ferritin genes (CaFer1 and CaFer2) were analyzed by Real-Time PCR. Al caused a downregulation of CaFER1 expression and no changes of CaFER2 expression in both varieties. The Western blot showed no alteration in ferritin protein levels in Mundo Novo and a decrease in Icatu. The differential enzymes responses indicate that the response to Al is variety-dependent.

    Caffeine Formation by Suspension Cultures of Coffea dewevrei

    The low caffeine content in leaves of C. dewevrei (~ 0.5 mg/g) is due to a low biosynthesis associated with a fast degradation. On the other hand, high biosynthesis and low degradation confer a higher content (~ 8 mg/g) in leaves of C. arabica. In this work it was observed that cell cultures of C. dewevrei recovered the ability to synthesize caffeine almost in similar levels of C. arabica cultures. Tracer experiments with labelled carbon dioxide showed a significant accumulation of radioactivity in caffeine and metabolites, indicating an active biosynthesis. When the cultures were fed with labelled caffeine most of the radioactivity was recovered in caffeine, indicating that although active, degradation was not so efficient as in leaves, and therefore, contributing for the alkaloid accumulation in the cell cultures.

    Brassinosteróides em café

    Testou-se o efeito da aspersão de soluções aquosas de 24-epibrassinotfdio e 24-epicastasterona, variando de 10-2 a 10-4ppm, em cafeeiros do cultivar Catuaí Amarelo, de Coffea arabica L. com dois anos de idade e na sua primeira produção. Avaliaram-se a produção de frutos, porcentagem de frutos com lojas vazias, porcentagem de sementes do tipo "moca", renda, peneira média, porcentagem de cada peneira (de 13 a 20) no peso total de sementes do tipo "chato" e, para apenas um experimento, o estabelecimento de frutos. Para os parâmetros em que ocorreram diferenças significativas, os tratamentos com brassinosteróídes nunca se mostraram estatisticamente superiores ao controle. Concluiu-se que os brassinosteróídes testados não afetaram a produção nem os outros parâmetros avaliados.
