    Drying of arabica coffee and its effect on the gene expression and activity of enzymes linked to seed physiological quality

    The reduced longevity of coffee seeds has been attributed to their sensitivity to desiccation. Studies related to gene expression and enzyme activity in coffee seeds under drying are important for understanding the effects of drying on their physiological quality. The aim of this study was to investigate the molecular aspects of seeds under different drying methods and associate them with physiological quality. Coffee seeds with different water contents were dried both slowly and rapidly. Enzymatic activity was analysed, as well as the expression of genes that encode the enzymes superoxide dismutase, catalase, peroxiredoxins, isocitrate lyase, and endo-ß-mannanase. There was a significant effect of drying speed and final water content on enzyme activity and on the expression of the different genes analysed. In seeds under rapid drying, there was greater expression of the genes that encode the enzymes catalase and endo-ßmannanase. Greater expression of the 1 CYS PRX and SOD genes and greater activity of the ICL isoenzymes were found in seeds with superior physiological quality, but greater activity of the endo-β-mannanase and CAT enzymes occurred in seeds with lower physiological quality.

    Alterações na qualidade do café submetido a diferentes formas de processamento e secagem

    Pesquisas recentes têm indicado várias alterações nos grãos de café decorrentes do processamento e secagem. Assim, neste trabalho objetivou-se verificar a influência da secagem lenta e secagem rápida sobre alguns parâmetros bioquímicos e fisiológicos de grãos de café e sobre a qualidade da bebida. Cafés da cultivar Catuaí Vermelho IAC 144 foram colhidos e submetidos a três formas de processamento, café natural, desmucilado e despolpado, e dois métodos de secagem, lenta à sombra em telados suspensos e secagem rápida em secadores de camada fixa a 35 ºC. Após secagem, os cafés foram submetidos às seguintes avaliações: análise sensorial, condutividade elétrica, lixiviação de potássio, atividade enzimática da polifenoloxidase, acidez total titulável e análises fisiológicas. Há interação entre métodos de processamento e de secagem sobre a qualidade sensorial do café, sendo mais evidentes os efeitos da secagem. Os cafés obtidos por via úmida tem maior tolerância à secagem do que os processados por via seca. Melhor qualidade sensorial é obtida nos cafés submetidos à secagem lenta, independentemente da forma de processamento utilizada. Cafés de melhor qualidade sensorial apresentam melhor qualidade fisiológica.

    Steps of cryopreservation of coffee seeds: physiological responses and antioxidant systems

    The cryopreservation of plant germplasm at ultralow temperatures is an alternative technique for the long-term storage of seeds of the genus Coffea sp. However, for this technique to be successful, cell integrity must be maintained at all stages of the process on the basis of scientific research. The present study investigated validated cryopreservation protocols for Coffea arabica L. seeds and evaluate the effects on the physiological and biochemical characteristics of the seeds at each stage of the process. Seeds were dried on silica gel or with saturated saline solution, precooled or not in a biofreezer, immersed in nitrogen, and reheated in a water bath. After each of these steps, the physiological and biochemical quality of the seeds was determined. Pre-cooling is a step that can be dispensed with in the cryopreservation of Coffea arabica seeds, direct immersion in liquid nitrogen being more indicated. Coffea arabica L. seeds tolerate cryopreservation after rapid drying in silica gel up to water contents of 17 or 20% (wb), with greater survival at 17%. The enzyme activities of catalase, polyphenol oxidase and peroxidase are indicators of the quality of C. arabica L. seeds subjected to cryopreservation.

    Evaluation of the behavior of coffee stored in cooled and natural environments

    The market value of coffee is strongly influenced by loss of quality, which makes storage one of the main steps in the entire production chain. The finite element method (FEM) and computational fluid dynamics (CFD) are numerical and computational techniques that facilitate the simulation of agricultural product storage systems. Computational modeling satisfactorily represents real experimentation, simplifies decision-making, and reduces costs. This study aimed to analyze mocha coffee storage for 6 months in a cooled environment with temperatures between 15 and 18 °C and in a natural environment. The water content, bulk density, specific heat, thermal conductivity, and thermal diffusivity were determined and colorimetry and sensory analysis were applied to compare initial and final samples of the product after storage. It was found that the water content and specific heat were the only properties that presented significant changes. Through sensory analysis, it was observed that the quality of the coffee was the same for both systems. A computational model was developed to simulate the heat transfer process during storage. The comparison of the simulation results with the experimental results for the temperature distribution in the grain mass showed overall mean relative errors of 2.34% for the natural environment and 5.74% for the cooled environment.

    Physiological quality of coffee seeds produced under different levels of solar radiation and maturation stages

    In Brazil, although the coffee plantations are predominantly grown under full sunlight, the use of agroforestry systems can lead to socioeconomic advantages, thus providing a favorable environment to the crop by promoting its sustainability as well as environmental preservation. However, there is a lack of information on physiological quality of the coffee seeds produced under different levels of solar radiation. Within this context, the objective of this study was to evaluate the influence of different levels of solar radiation and maturation stages on the physiological quality of coffee (Coffea arabica L.) seeds, cv. Acaiá Cerrado MG-1474. Three levels of solar radiation (plants grown under full sunlight; under plastic screens of 35% shading; and under plastic screens of 50% shading) and three maturation stages (cherry, greenish-yellow and green) were assessed. Physiological quality of seeds was assessed by using germination test, first count of germination, abnormal seedlings, dead seeds, and seedlings with open cotyledonary leaves. Electrophoretic analysis of isoenzymes catalase, esterase, superoxide dismutase and peroxidase was also performed. With the evolution of development the coffee seeds presents increases on physiological quality, and at its beginning the seeds show improvements on quality with the reduction of solar radiation.

    Armazenabilidade de sementes de cafeeiro colhidas em diferentes estádios de maturação e submetidas a diferentes métodos de secagem

    O momento da colheita e os métodos de secagem podem influenciar a qualidade das sementes de cafeeiro durante o armazenamento. Este trabalho foi realizado com o objetivo de avaliar os efeitos do estádio de maturação e do método de secagem sobre a qualidade fisiológica e a armazenabilidade de sementes de cafeeiro. Os ensaios foram realizados nos Laboratórios de Análise de Sementes e de Técnicas Moleculares do Departamento de Agricultura da UFLA. Os frutos do cultivar Rubi foram colhidos, despolpados, e as sementes lavadas e deixadas sobre papel para retirada da água superficial. As sementes nos estádios verde cana e cereja foram submetidas à secagem convencional (à sombra) e à secagem em secador estacionário sob temperatura de 35ºC. Como testemunha foram analisadas sementes sem secagem. As avaliações foram feitas imediatamente após os tratamentos de secagem e após 4 e 8 meses de armazenamento. As sementes foram armazenadas a 10ºC em sacos plásticos impermeáveis. Foram realizados os testes de germinação, de protrusão radicular, de matéria seca de plântulas, de índice de velocidade de emergência, de condutividade elétrica, além de análises eletroforéticas de proteínas resistentes ao calor e da quantificação da atividade da enzima endo-ß-mananase. O delineamento foi inteiramente ao acaso em esquema fatorial dois (estádios de maturação) x três (métodos de secagem) x três (tempos de armazenamento), com quatro repetições. Sementes de cafeeiro colhidas no estádio cereja têm maior potencial de armazenamento que no estádio verde cana. Ocorre redução de germinação e vigor nas sementes de cafeeiro colhidas no estádio verde cana, quando submetidas à secagem rápida. A presença ou intensidade de bandas de proteínas resistentes ao calor está associada à secagem das sementes. Ocorre maior atividade da enzima endo-ß-mananase em sementes colhidas no estádio cereja que no estádio verde-cana. Ocorre aumento da atividade da enzima endo-ß-mananase durante o armazenamento.

    Inibição do desenvolvimento in vitro de embriões de Coffea por cafeína exógena

    A cafeína, alcalóide conhecido como 1,3,7-trimetilxantina, é encontrada em sementes quiescentes de cafeeiro, perfazendo um total de 1,1 a 1,7% em Coffea arabica L. e 2 a 3% em Coffea canephora Pierre, localizada em sua grande maioria no endosperma, na forma livre no citoplasma das células ou complexada com ácidos clorogênicos. Com função fisiológica em plantas ainda não totalmente esclarecida, a cafeína causa efeito alelopático, seja inibindo a germinação de várias espécies, seja como anti-herbívoro ou como agente pesticida natural. A lenta germinação de sementes de café ainda não foi esclarecida e várias causas são apontadas, como presença do endocarpo, baixa absorção de água e O2, presença de inibidores naturais e balanço hormonal. Embora sugeridos, estudos sobre a inibição de sementes de cafeeiro por ação da cafeína endógena e ou exógena são escassos. Assim, o presente trabalho teve como objetivo estudar o efeito da cafeína exógena sobre a germinação e o desenvolvimento de embriões de Coffea arabica L. e de Coffea canephora Pierre. O experimento foi realizado utilizando-se frutos no estádio cereja das cultivares Rubi e Apoatã IAC-2258. Após desinfestação dos frutos por 30 minutos de imersão em hipoclorito de sódio (2% i.a.) e lavagem por três vezes em água destilada e autoclavada, os embriões foram retirados e inoculados, de modo asséptico, em placas de petri com meio MS 50%, acrescido de sacarose e suplementado com diferentes concentrações de cafeína (0,00; 0,05; 0,10; 0,15; 0,20; 0,25; 0,30 e 0,40%). Os embriões foram mantidos em sala de crescimento a 27 ± 2ºC e densidade de fluxo de fótons de 13µmol.m-2.s-1, durante 23 dias, quando foram avaliados comprimento da parte aérea, comprimento de raiz e massa fresca das plântulas. Cinco dias após o cultivo, foram avaliadas a porcentagem de emissão de radículas e cotilédones, computando-se os embriões com cotilédones abertos e radículas expandidas. O delineamento experimental utilizado foi inteiramente casualizado com seis repetições por tratamento, sendo cada repetição constituída por cinco embriões. Concluiu-se que a germinação e o desenvolvimento in vitro de embriões de Coffea arabica L. e Coffea canephora Pierre são afetados por cafeína exógena. O efeito detrimental da cafeína exógena em embriões de Coffea é maior nas radículas do que nos cotilédones. Embriões de Coffea arabica L. são mais sensíveis aos efeitos negativos da cafeína exógena do que embriões de Coffea canephora Pierre. A cafeína pode contribuir para a lenta germinação de sementes e o lento desenvolvimento de plântulas de cafeeiro.

    Protein profile in arabica coffee seeds in electrophoresis gel: importance of freeze-drying

    Coffee seeds are sensitive to desiccation and are used or stored with different moisture content values, which may affect the results of quality assessment. The aim of this study was to evaluate the changes in protein profile in electrophoresis gel in coffee seeds with different moisture content values under freeze-drying and without freeze-drying. Two lots of arabica coffee seeds were used, one of newly-harvested seeds and another of stored seeds. The seeds were dried to the moisture content values of 12, 15, 20, 25, 30, 35, and 40%. The physiological quality of the seeds was assessed through the germination test, electrical conductivity, and the profiles of the enzymes SOD, CAT, PO, GOT, MDH, and EST and of heat-resistant proteins. In general, there is an effect on expression of these enzymes in accordance with the presence of free water in the seeds. Moist seeds have little to no enzyme expression. The freeze-drying process allows preservation of coffee seed quality and does not change the functionality of the enzymes studied. The isoenzyme profiles of the antioxidant process in arabica coffee seeds are affected by the initial moisture content of the seeds. The freeze-drying process of the seeds ensures greater sensitivity in detection of the expression of these isoenzymes.

    Trolox equivalent antioxidant capacity of Coffea arabica L. seeds

    The causes of the low desiccation tolerance and low longevity of coffee seeds have not yet been fully elucidated, and a full understanding of their complex physiology is of great interest. Among several alternatives, the loss of antioxidant capacity in seeds may be related to their rapid loss in quality during storage. The objective of this study was to determine the total antioxidant capacity of coffee harvested at different ripeness stages before and after the storage of seeds with different water contents and to relate antioxidant capacity to physiological quality. Seeds in the greenish-yellow or cherry stages, recently harvested or stored for nine months at 10 °C with 40, 30, 20 and 12% water content (wet basis - wb), were submitted to physiological and biochemical quality evaluations, and the Trolox equivalent antioxidant capacity (TEAC) was determined. The germination and root protrusion of coffee seeds from greenish-yellow and cherry fruits were not affected by drying, but seeds harvested at physiological maturity had greater vigor when the moisture content was lower. The quality of coffee seeds decreased during storage, and this decrease was greater in seeds stored with higher water contents. Coffee seeds in the greenish-yellow stage had a higher antioxidant capacity than those in the cherry stage when recently harvested, but there was a substantial reduction in this capacity during storage at both maturation stages. Coffee seed deterioration is related to a reduction in antioxidant capacity, and the isoenzymatic profiles of the antioxidant process are little affected by the seed maturation stage. The deterioration of coffee seeds during storage is related to a reduction in their total antioxidant capacity, regardless of their maturation stage, being more pronounced in the greenish-yellow stage

    Desiccation sensitivity from different coffee seed phenological stages

    Maturity stage and drying method are the factors that most influence coffee seed quality. The objective of this study was to assess the physiological quality and investigate the electrophoretic patterns of catalase and endo-ß-mannanase enzymes and heat resistant proteins in coffee seeds harvested at different phenological stages and dried under different conditions. Physiological quality was assessed when the seeds had developed the green, greenish-yellow, cherry, overripe and dry stages after three treatments: no drying, conventional drying and fast drying. After each treatment, the physiological quality of the seeds was assessed using the germination test and electrophoretic patterns of heat resistant proteins and the activity of catalase and endo-ß-mannanase enzymes. Seeds harvested at the cherry phenological stage had the best physiological quality, and the drying process reduced quality at the cherry, overripe and dry stages. This reduction was greater under the faster drying process, but at the greenish-yellow stage, seeds had better physiological quality after slow drying. Regarding the results from electrophoretic analysis, endo-ß-mannanase and catalase activities increase as the ripeness stages advance; the activity of endo-ß-mannanase is directly associated with the deterioration process; the expression of heat resistant proteins increases with maturation process and is associated with seed physiological quality.
