However, there is a lack of information on physiological quality of the coffee seeds produced under different levels of solar radiation. Within this context, the objective of this study was to evaluate the influence of different levels of solar radiation and maturation stages on the physiological quality of coffee (Coffea arabica L.) seeds, cv. Acaiá Cerrado MG-1474. Three levels of solar radiation (plants grown under full sunlight; under plastic screens of 35% shading; and under plastic screens of 50% shading) and three maturation stages (cherry, greenish-yellow and green) were assessed. Physiological quality of seeds was assessed by using germination test, first count of germination, abnormal seedlings, dead seeds, and seedlings with open cotyledonary leaves. Electrophoretic analysis of isoenzymes catalase, esterase, superoxide dismutase and peroxidase was also performed. With the evolution of development the coffee seeds presents increases on physiological quality, and at its beginning the seeds show improvements on quality with the reduction of solar radiation.Item Inibição do desenvolvimento in vitro de embriões de Coffea por cafeína exógena(Associação Brasileira de Tecnologia de Sementes, 2006-05-30) Rosa, Sttela Dellyzete Veiga Franco da; Santos, Cíntia Guimarães dos; Paiva, Renato; Melo, Patrícia Leonardo Queiroz de; Veiga, André Delly; Veiga, Adriano DellyA cafeína, alcalóide conhecido como 1,3,7-trimetilxantina, é encontrada em sementes quiescentes de cafeeiro, perfazendo um total de 1,1 a 1,7% em Coffea arabica L. e 2 a 3% em Coffea canephora Pierre, localizada em sua grande maioria no endosperma, na forma livre no citoplasma das células ou complexada com ácidos clorogênicos. Com função fisiológica em plantas ainda não totalmente esclarecida, a cafeína causa efeito alelopático, seja inibindo a germinação de várias espécies, seja como anti-herbívoro ou como agente pesticida natural. A lenta germinação de sementes de café ainda não foi esclarecida e várias causas são apontadas, como presença do endocarpo, baixa absorção de água e O2, presença de inibidores naturais e balanço hormonal. Embora sugeridos, estudos sobre a inibição de sementes de cafeeiro por ação da cafeína endógena e ou exógena são escassos. Assim, o presente trabalho teve como objetivo estudar o efeito da cafeína exógena sobre a germinação e o desenvolvimento de embriões de Coffea arabica L. e de Coffea canephora Pierre. O experimento foi realizado utilizando-se frutos no estádio cereja das cultivares Rubi e Apoatã IAC-2258. Após desinfestação dos frutos por 30 minutos de imersão em hipoclorito de sódio (2% i.a.) e lavagem por três vezes em água destilada e autoclavada, os embriões foram retirados e inoculados, de modo asséptico, em placas de petri com meio MS 50%, acrescido de sacarose e suplementado com diferentes concentrações de cafeína (0,00; 0,05; 0,10; 0,15; 0,20; 0,25; 0,30 e 0,40%). Os embriões foram mantidos em sala de crescimento a 27 ± 2ºC e densidade de fluxo de fótons de 13µmol.m-2.s-1, durante 23 dias, quando foram avaliados comprimento da parte aérea, comprimento de raiz e massa fresca das plântulas. Cinco dias após o cultivo, foram avaliadas a porcentagem de emissão de radículas e cotilédones, computando-se os embriões com cotilédones abertos e radículas expandidas. O delineamento experimental utilizado foi inteiramente casualizado com seis repetições por tratamento, sendo cada repetição constituída por cinco embriões. Concluiu-se que a germinação e o desenvolvimento in vitro de embriões de Coffea arabica L. e Coffea canephora Pierre são afetados por cafeína exógena. O efeito detrimental da cafeína exógena em embriões de Coffea é maior nas radículas do que nos cotilédones. Embriões de Coffea arabica L. são mais sensíveis aos efeitos negativos da cafeína exógena do que embriões de Coffea canephora Pierre. A cafeína pode contribuir para a lenta germinação de sementes e o lento desenvolvimento de plântulas de cafeeiro.Item Desiccation sensitivity from different coffee seed phenological stages(Associação Brasileira de Tecnologia de Sementes - ABRATES, 2014) Santos, Flávia Carvalho; Rosa, Sttela Dellyzete Veiga Franco da; Pinho, Édila Vilela de Rezende Von; Cirillo, Marcelo Angelo; Clemente, Aline da Consolação SampaioMaturity stage and drying method are the factors that most influence coffee seed quality. The objective of this study was to assess the physiological quality and investigate the electrophoretic patterns of catalase and endo-ß-mannanase enzymes and heat resistant proteins in coffee seeds harvested at different phenological stages and dried under different conditions. Physiological quality was assessed when the seeds had developed the green, greenish-yellow, cherry, overripe and dry stages after three treatments: no drying, conventional drying and fast drying. After each treatment, the physiological quality of the seeds was assessed using the germination test and electrophoretic patterns of heat resistant proteins and the activity of catalase and endo-ß-mannanase enzymes. Seeds harvested at the cherry phenological stage had the best physiological quality, and the drying process reduced quality at the cherry, overripe and dry stages. This reduction was greater under the faster drying process, but at the greenish-yellow stage, seeds had better physiological quality after slow drying. Regarding the results from electrophoretic analysis, endo-ß-mannanase and catalase activities increase as the ripeness stages advance; the activity of endo-ß-mannanase is directly associated with the deterioration process; the expression of heat resistant proteins increases with maturation process and is associated with seed physiological quality.Item Exploratory studies for cryopreservation of Coffea arabica L. seeds(Associação Brasileira de Tecnologia de Sementes - ABRATES, 2017) Figueiredo, Madeleine Alves de; Coelho, Stefania Vilas Boas; Rosa, Sttela Dellyzete Veiga Franco da; Vilela, Amanda Lima; Silva, Luciano CoutinhoCryopreservation is a viable option for conservation of coffee germplasm. However, for this technique to be completely successful, it is of fundamental importance to carry out studies that ensure maintenance of cell integrity before and after immersion in liquid nitrogen (LN). Therefore, the aim in this study was to investigate the water content, cooling rate, and final temperature most suitable for cryopreservation of Coffea arabica L. seeds. The seeds were dried by silica gel to water contents of 5, 10, 15, 20, 30 and 40 % wb, subjected to slow cooling treatments at speeds of -1, -3 and -5 °C min.-1 to final temperatures of -40, -50 and -60 °C and then directly immersed in LN. After storage, seeds were rewarmed at 40 °C for two minutes. The survival rate and viability of the seeds and embryos were evaluated by the tetrazolium and germination tests. Results of the tetrazolium test indicate that embryos excised from cryopreserved seeds are less sensitive to cryopreservation than whole seeds are. The water content of 20% wb and the use of zygotic embryos led to the highest survival rate of the coffee seeds, depending on the cooling rate and the final temperature of precooling.Item Association between the artificial aging test and the natural storage of coffee seeds(Associação Brasileira de Tecnologia de Sementes - ABRATES, 2018) Fantazzini, Tatiana Botelho; Rosa, Sttela Dellyzete Veiga Franco da; Pereira, Cristiane Carvalho; Pereira, Diego de Sousa; Cirillo, Marcelo Ângelo; Ossani, Paulo CésarThe accelerated aging test is recognized as an efficient method for evaluating the vigor of seed lots and for estimating their storage potential. Thus, this work aimed to evaluate the association between artificial aging and natural storage of coffee seeds, through the correlation factor analysis. Seeds of four cultivars of Coffea arabica L. (Catuaí Amarelo, Arara, Catiguá, and Mundo Novo) and one of Coffea canephora Pierre (Apoatã) were used. Part of the newly-harvested seeds were aged in a growth chamber under controlled temperature and relative humidity conditions (42 ºC and 100% RH) for periods of 0, 4, 6, 8, and 10 days. The other part of the seeds was stored in tri-wall paper packaging for a period of 2, 4, and 6 months in a non-climate-controlled environment. The aim of this study was to investigate the antioxidant and antimicrobial activity of cathode and anode water on the physiological quality and health of Coffea arabica L. seeds. Part of the seeds were soaked in cathode water and the other part in anode water for periods of 1, 6, 12, 24, and 36 hours. The physiological quality of the seeds was evaluated by the germination test and tetrazolium test, and seed health by the Blotter test. Analysis of variance and Pearson correlation analysis were performed on the data for all the response variables. Soaking in cathode water or anode water up to 24 hours improves the physiological quality of coffee seeds. Anode water has a beneficial effect on the health of coffee seeds, reducing the incidence of Fusarium spp., Penicillium spp., Cladosporium spp., and Alternaria spp. after one hour of soaking and of Aspergillus spp. after six hours. Soaking in cathode water for a period of one hour leads to a reduction in the fungi Aspergillus spp., Fusarium spp., Cladosporium spp., and Alternaria spp., and reduction in Penicillium spp. after six hours.Item Physiological, biochemical, and ultrastructural aspects of Coffea arabica L. seeds under different cryopreservation protocols(Editora UFLA, 2021) Figueiredo, Madeleine Alves de; Rosa, Sttela Dellyzete Veiga Franco da; Ricaldoni, Marcela Andreotti; Pereira, Cristiane Carvalho; Coelho, Stefânia Vilas Boas; Silva, Luciano CoutinhoCryopreservation is a technique that may potentially conserve the germplasm of species of the Coffea genus for an indeterminate time. The aim of this study was to evaluate the physiological, biochemical and ultrastructural characteristics of cryopreserved seeds of Coffea arabica L., cultivar Catucaí amarelo IAC 62, which was subjected to different protocols regarding dehydration, precooling, cooling, rewarming and cathode water use. According to each protocol, the seeds were subjected to fast or slow drying to moisture contents of 17 or 20% (wet basis), cooled in different ways, and then immersed in liquid nitrogen for 24 hours. Different rewarming times in a water bath were also used. Physiological, biochemical and ultrastructural analyses were performed on the seeds after the cryopreservation steps. Moisture content at a 17% wb is the key factor for the cryopreservation of Coffea arabica L. seeds, which have better physiological quality and better preserved cell structures. Precooling of coffee seeds before immersion in liquid nitrogen does not provide advantages compared to direct immersion. The rewarming times tested (2, 4, and 6 minutes) and cathode water use did not cause changes in the physiological and biochemical quality or in the cell structures of Coffea arabica L. cryopreserved seeds. The pattern of cell structure observed in all seeds indicates that the damage from cryopreservation is less drastic in the cells of the embryos than in those of the endosperm, with the latter less tolerant to the stresses of dehydration, precooling, and rewarming.Item Ultrastructural damage in coffee seeds exposed to drying and to subzero (°C) temperatures(Editora UFLA, 2020) Coelho, Stefânia Vilas Boas; Rosa, Sttela Dellyzete Veiga Franco da; Clemente, Aline da Consolação Sampaio; Lacerda, Laura Nardelli Castanheira; Silva, Luciano Coutinho; Fantazzini, Tatiana Botelho; Ribeiro, Fernando Augusto Sales; Castro, Elisa de MeloDuring drying and freezing, protective mechanisms act to maintain seed physiological quality. Some of these mechanisms contribute to the integrity of cell membranes. The damage caused to cell membranes due to cell stress can be seen in ultrastructural studies, comparing these results to those of physiological evaluation. The aim of this study was to investigate ultrastructural changes in endosperm cells of coffee seeds brought about by drying and by exposure to low temperatures. Seeds of Coffea arabica were dried in silica gel to moisture contents of 40, 20, and 5 % (wb) and brought to equilibrium at temperatures of 10, -20, and -86 oC. Germination, vigor, and tetrazolium tests were performed for evaluation of seed physiological quality. Ultrastructural damage was analyzed by scanning electron microscopy. Coffee seeds with 40% moisture content have whole, swollen, and expanded cells, with a filled lumen and without signs of damage. The physiological and ultrastructural quality of seeds exposed to below zero temperatures with 40% moisture content is compromised. They have null germination and empty cells, indicating leakage of cell content. Drying of coffee seeds leads to uniform contraction of inner cell content. Drying of coffee seeds to 5% moisture content leads to intense contraction of cell volume, with physiological and ultrastructural damage.Item Cryopreservation in Coffea canephora Pierre seeds: slow and fast cooling(Editora UFLA, 2018-11) Coelho, Stefânia Vilas Boas; Rosa, Sttela Dellyzete Veiga Franco da; Fantazzini, Tatiana Botelho; Baute, Júlia Lima; Silva, Luciano CoutinhoCoffee is one of the main agricultural commodities in the country, and it is important to conservation of plant material for breeding programs. Cryopreservation is a promising alternative for preserving in the long-term the germplasm of species considered recalcitrant. However, studies should be performed to achieve maximum survival of seedlings after immersion in liquid nitrogen. The objective of this work was to find a cryopreservation protocol for storing seeds of Coffea canephora, studying two methods of cryopreservation, slow and fast cooling. Seeds were subjected to drying in silica gel up to the water content of 0.25 g g -1 . In the first experiment, dried seeds were subjected to treatments of slow cooling at speeds of -1 oC min -1 ,-3 oC min -1 and -5 oC min -1 until the end temperatures of -40 oC, -50 oC and -60 oC, by means of a bio freezer and subsequently immersed in liquid nitrogen. In the second experiment, the best result was selected of the first experiment and compared with the rapid cooling, in which dried seeds, with 0.25 g g -1 of water content, were immersed directly into liquid nitrogen. Physiological and biochemical alterations occurring in the seeds after cryopreservation were evaluated. Coffea canephora seeds respond better to cryopreservation by rapid cooling, when compared to slow cooling. Drying, one of the cryopreservation steps does not affect the viability of Coffea canephora Pierre seeds, when these seeds are dried to 0.25 g g -1 of water content.
