    Sprouting induction for micro-cutting on in vitro cloned arabica coffee plants

    Vegetative propagation of arabica coffee plants selected by their agronomic value has been accomplished routinely in Brazil for scientific purposes, through somatic embryogenesis and rooting of stem cuttings. Somatic embryogenesis is the election method when a very high number of cloned plants is demanded. Nevertheless, the costs of in vitro multiplication make difficult to explore it commercially. The experiments described herein aimed to amplify the number of in vitro cloned plants, post acclimatization, to reduce costs. Different concentrations of 2,3,5-triiodobenzoic acid (TIBA) and its association with benzylaminopurine (BAP) were applied, as successive pulses, in the 3 rd , 8 th and 13 th months after transference to the greenhouse, on the same set of Catucaí and Siriema in vitro cloned plants, to induce sprouting. At the 8 th month, the experiments with in vitro cloned Catucaí plants were reproduced in the nursery, for comparison. Best results were observed for the association TIBA 1000 mg.mL-1 + BAP 60 mg.mL-1 applied in the greenhouse, at the 13 th month, when, on average, 8.5 and 7.0 micro-cuttings above 1 cm in length were produced using sprouts taken from each Catucaí and Siriema acclimatized plant, respectively. Applying this treatment twice a year, and harvesting induced sprouts each six months after the induction treatments, approximately 15 plants per each acclimatized one can be produced. The most important effect of TIBA was the induction of sub-apical sprouting. Greenhouse would be the best environment to apply successive pulses of sprouting inducers to coffee in vitro cloned plants.

    Effect of 6-BA on nodal explant bud sproutings of Coffea arabica cv. Mundo Novo

    Coffee plants can be micropropagated by nodal bud sprouting using the 6-benzylaminopurine (6-BA) hormone. However, literature reports the use of a wide range of 6-BA, from 0.5 to 88.8 µM L-1. So, this study was performed to narrow that range. Nodal explants of Coffea arabica cv Mundo Novo obtained from in vitro plantlets were inoculated on gelled-MS medium supplemented with different concentrations of 6-BA. Two assays were carried out: in the first one, 6-BA was used at concentrations of 0, 5, 25, 50, and 100 µM L-1, being evaluated at 43 and 123 days. In the second experiment, dosis of 10, 20 and 30 µM L-1, have evaluated at 65 and 100 days. Treatments with 6-BA induced multiple sprouting from the nodal explants, which were best characterized around 100 days after inoculation. The nodal explants grew taller and showed multiple shoots, whereas the effect of 6-BA at 5 to 25 µM L-1 was similar to that with higher concentrations (50 and 100 µM L-1). Nodal explants yielded from 2.9 to 6.0 buds per node, achieving height of 1.3 to 1.5 cm at 5 to 25 µM L-1 of 6-BA, whereas they yielded from 4.3 to 4.9 buds per node but the sprouting grew about 0.8 cm at 50 and 100 µM L-1 of 6-BA. This study indicated that multiple sprouting of lateral buds can be induced by lower concentrations of 6-BA, for example, from 10 to 30 µM L-1, diminishing possible risks of somaclonal variation due to high levels of hormone concentration.

    Development of Coffea arabica L. seedling obtained from direct somatic embryogenesis

    This work aimed to develop embryos and acclimatize seedlings of Coffea arabica L. cv. Rubi produced by direct somatic embryogenesis. For the development of somatic embryos, were evaluated the effect of sucrose (0; 15; 30; 45 and 60 g.L-1) and GA3 (0; 2.5; 5 and 10 mg.L-1). For the development of seedlings, were evaluated the influence of GA3 (0; 2.5; 5; 10 mg.L-1) and NAA (0; 0.25; 0.5; 1; 2 mg L-1). The in vitro experiments were carried out in a growth chamber under light intensity of 32 μMol.m-2.s-1, at the temperature of ± 25 1oC and photoperiod of 16 hours. The parameters evaluated were: number of leaves, length of the aerial part and fresh weight of the seedlings. For the seedling acclimatization process two types of substrates were tested (Plantmax® and Plantmax® plus carbonized rice peels (v/v 1:1) and fertilizer with slow-release (Osmocote®) (0, 0.5, 1 and 2 g of the fertilizer per cell). The results indicated that 5.81 mg L-1 GA3 was efficient for the development of coffee somatic embryos and GA3 (10 mg. L-1) in combination with 1mg. L-1 NAA showed efficiency in the development of the seedlings obtained by direct somatic embryogenesis. The best substrate for acclimatization was Plantmax® mixed with carbonized rice peels and 1.68 g per cell of the Osmocote® fertilizer.

    Influência de reguladores de crescimento no desenvolvimento radicular de sementes de Coffea arabica L. 'Rubi' in vitro

    Avaliou-se o efeito de reguladores de crescimento na germinação e enraizamento in vitro de sementes de cafeeiro Rubi (Coffea arabica L. Rubi ). Sementes de plantas adultas de cafeeiro foram inoculadas em meio MS/2 + 20 g L-1 de sacarose e 6 g L-1 de ágar. Foram testados quatro tratamentos: T1 - controle; T2 - 6 mg L-1 de BAP + 0,1 mg L-1 de ANA; T3- 6 mg L-1 de BAP; T4 - 5 mg L-1 de GA3. O melhor tratamento obtido para enraizamento de sementes de cafeeiro foi o controle (MS/2 + ágar + sacarose), tornandose desnecessária a adição de reguladores de crescimento.
