    Inhibition of Candida albicans growth by steeping freeze-dried robusta ground coffee

    Coffee contains active compounds that alleged to have antifungal activity. This study aimed to analyze inhibitory effect of steeping freeze–dried Robusta ground coffee (SFDRGC) to the growth of oral microorganism Candida albicans (the main causative agent of oro-pharyngeal opportunistic infection). This is an in vitro experimental study using the post–test only control group design. Serial dilutions of SFDRGC were studied. An antifungal drug Nystatin was used as a positive control, and sterile distilled water as a negative control. The inhibitory effect was study by means of Agar well diffusion method. Result showed that SFDRGC demonstrated antifungal activity. The higher concentration of SFDRGC showed the higher antifungal activities significantly (P < 0.05). Concentration of 100% SFDRGC showed anti-candida activity as potent as Nystatin. In conclusion, 100% SFDRGC demonstrated a potent antifungal agent against C. albicans. This study suggested that coffee beverage might be used as anti-candida mouthwash to prevent the risk of oral opportunistic infection. Further studies, however are needed to confirm this notion, such as epidemiological, and phytochemical studies of antifungal bioactive components in coffee that are processed using various methods.

    Sprouting induction for micro-cutting on in vitro cloned arabica coffee plants

    Vegetative propagation of arabica coffee plants selected by their agronomic value has been accomplished routinely in Brazil for scientific purposes, through somatic embryogenesis and rooting of stem cuttings. Somatic embryogenesis is the election method when a very high number of cloned plants is demanded. Nevertheless, the costs of in vitro multiplication make difficult to explore it commercially. The experiments described herein aimed to amplify the number of in vitro cloned plants, post acclimatization, to reduce costs. Different concentrations of 2,3,5-triiodobenzoic acid (TIBA) and its association with benzylaminopurine (BAP) were applied, as successive pulses, in the 3 rd , 8 th and 13 th months after transference to the greenhouse, on the same set of Catucaí and Siriema in vitro cloned plants, to induce sprouting. At the 8 th month, the experiments with in vitro cloned Catucaí plants were reproduced in the nursery, for comparison. Best results were observed for the association TIBA 1000 mg.mL-1 + BAP 60 mg.mL-1 applied in the greenhouse, at the 13 th month, when, on average, 8.5 and 7.0 micro-cuttings above 1 cm in length were produced using sprouts taken from each Catucaí and Siriema acclimatized plant, respectively. Applying this treatment twice a year, and harvesting induced sprouts each six months after the induction treatments, approximately 15 plants per each acclimatized one can be produced. The most important effect of TIBA was the induction of sub-apical sprouting. Greenhouse would be the best environment to apply successive pulses of sprouting inducers to coffee in vitro cloned plants.

    Tolerância ao déficit hídrico de cafeeiros produzidos por estaquia e embriogênese somática

    A irrigação em lavouras cafeeiras é foco de pesquisas há muitos anos, porém as novas tecnologias de produção de mudas (embriogênese somática e estaquia) têm demandado novos estudos de comportamento dessas em campo e suas necessidades hídricas. Um experimento foi conduzido em casa de vegetação, na Universidade Federal de Lavras, Lavras – MG, objetivando-se avaliar a tolerância ao déficit hídrico de cafeeiros oriundos de mudas obtidas por embriogênese somática e estaquia, na fase de implantação da cultura. O delineamento utilizado foi o de blocos casualizados, com cinco repetições, esquema fatorial 2 x 5 sendo dois tipos de mudas, produzidas por embriogênese somática e estaquia, além de cinco níveis de irrigação, baseados na manutenção da capacidade de campo (CC) do solo a 20%, 40%, 60%, 80% e 100% da CC. Aos 153 dias após o início do experimento foram avaliados os teores foliares de prolina, potencial hídrico foliar, altura de plantas, diâmetro de caule, área foliar, peso seco de raízes e parte aérea. Constatou-se que cafeeiros oriundos de estaquia e embriogênese somática respondem positivamente e progressivamente à irrigação. Verificou-se também que a capacidade de campo do solo mantida abaixo de 74,6% e 100%, para cafeeiros oriundos de mudas de embriogênese somática e estaquia respectivamente, prejudicam o crescimento e desenvolvimento iniciais das plantas.

    Vegetative vigor conilon coffee and its potential for in vitro callus induction

    Considering the importance of the culture of Coffea canephora in the Brazilian Amazon and the need for genetic improvement of this species for cultivation in the state of Rondonia, he objective of this study was to evaluate the correlation between the initial vigor of clonal plants of C. canephora variety Conilon, under field conditions, and its potential for in vitro propagation. Promising clones belonging to the Genetic Improvement Program of Coffea sp were used Embrapa (Brazilian Agricultural Research Corporation) Rondonia. The plant height, number and length of orthotropic shoots in coffee at 10 months after planting were evaluated. In the laboratory, leaves of plants of each clone were segmented into fragments of 1 cm 2 which were inoculated on MS medium containing half the concentration of salts, AIB (10 μM), 2,4-D (20 μM) and 2iP (10 μM), sugar (20 g cm -3 ) and agar (6 g cm -3 ). Cultures were maintained in a growth chamber under 16 h photoperiod at 2000 lux and 24 ± 2 o C. The callus induction and the percentage of leaf area covered with callus cells were observed within 90 days. Clone 9 showed greater vegetative growth in the field. Only clone 12 showed no callus induced, all other clones were highly responsive to callus induction, and clone 5 showed a higher percentage of leaf area covered with callus cells. There was no phenotypic correlation between early vigor in the field and calogênico potential in vitro, for clones of coffee used.

    Pigments, total soluble phenols and lignin levels of coffee seedlings inoculated with Colletotrichum gloeosporioides

    The objective of this work was to study the effects of different isolates of Colletotrichum gloeosporioides, as to pigment level (chlorophyll a, b and total), total soluble phenols and soluble lignin in coffee seedlings obtained by artificially inoculated tissue culture. The experiment was carried out in randomized block design in a 2 x 3 factorial scheme (2 genetic materials), 3 isolates (an isolate from mango fruits and another one from coffee plants), with 5 replicates and control (plants without inoculation). The plants were inoculated with a suspension of 2 x 10 6 conidia.ml -1 . The conidial suspension was sprayed on previously injured leaves. Leaves of inoculated plants were collected at 3 and 7 days after inoculation, except for leaves on the lower part of the plants. Afterwards, samples were prepared for the evaluation of chlorophyll, total soluble phenols and soluble lignin. The results showed that the presence of the pathogen in healthy and diseased plants significantly reduced the amount of chlorophyll. At 7 days after inoculation of healthy and diseased coffee plants higher total soluble phenol and soluble lignin levels were observed in response to the attack by Colletotrichum gloeosporioides.

    Comportamento de cafeeiros propagados por embriogênese somática e por sementes em diferentes níveis de água no solo

    A propagação vegetativa do cafeeiro via embriogênese somática (ES) é uma ferramenta bastante útil em programas de melhoramento genético e para a produção de mudas clonais em larga escala. Essa técnica permite a multiplicação de híbridos e de genótipos segregantes, de alto valor agronômico e possibilita reduzir, consideravelmente, o tempo necessário para o lançamento de novas cultivares. Todavia, há poucas informações sobre o comportamento de plantas obtidas por ES sob condições de baixa disponibilidade de água no solo. Nesse contexto, objetivou-se avaliar o efeito de diferentes níveis de água no solo em cafeeiros propagados via embriogênese somática, em comparação com cafeeiros propagados por sementes. Instalou-se um ensaio, em casa de vegetação, em esquema fatorial 4x2, sendo quatro níveis de água disponível no solo e dois métodos de propagação (embriogênese somática e semente), utilizando-se mudas de seis meses de idade de café arábica (Coffea arabica L.) da cultivar Catuaí Vermelho IAC 44. Foram avaliados a massa seca das folhas, do caule e das raízes, a área foliar, a altura, o diâmetro do caule, o comprimento radicular e o número de nós do ramo ortotrópico. Concluiu-se que plantas propagadas por embriogênese somática apresentam desenvolvimento semelhante ao de plantas obtidas por sementes em diferentes condições de disponibilidade de água no solo.

    Embriogênese somática em genótipos de Coffea arabica L.

    A espécie Coffea arabica L. possui genótipos com características de interesse, como o AC1, AC2 e AC3, cujas sementes apresentam conteúdo reduzido de cafeína. No entanto, cada um desses genótipos conta com uma única planta no Banco de Germoplasma do IAC, tendo, portanto, somente um acesso. Objetivou-se com este estudo avaliar a capacidade de embriogênese somática indireta dos genótipos AC e da cultivar Mundo Novo visando à sua multiplicação. Verificou-se que os três genótipos apresentaram capacidade embriogênica e formaram calos de tamanhos semelhantes entre si; no entanto, quanto à formação de embriões, o AC1 foi o mais eficiente.

    Development of Coffea arabica L. seedling obtained from direct somatic embryogenesis

    This work aimed to develop embryos and acclimatize seedlings of Coffea arabica L. cv. Rubi produced by direct somatic embryogenesis. For the development of somatic embryos, were evaluated the effect of sucrose (0; 15; 30; 45 and 60 g.L-1) and GA3 (0; 2.5; 5 and 10 mg.L-1). For the development of seedlings, were evaluated the influence of GA3 (0; 2.5; 5; 10 mg.L-1) and NAA (0; 0.25; 0.5; 1; 2 mg L-1). The in vitro experiments were carried out in a growth chamber under light intensity of 32 μMol.m-2.s-1, at the temperature of ± 25 1oC and photoperiod of 16 hours. The parameters evaluated were: number of leaves, length of the aerial part and fresh weight of the seedlings. For the seedling acclimatization process two types of substrates were tested (Plantmax® and Plantmax® plus carbonized rice peels (v/v 1:1) and fertilizer with slow-release (Osmocote®) (0, 0.5, 1 and 2 g of the fertilizer per cell). The results indicated that 5.81 mg L-1 GA3 was efficient for the development of coffee somatic embryos and GA3 (10 mg. L-1) in combination with 1mg. L-1 NAA showed efficiency in the development of the seedlings obtained by direct somatic embryogenesis. The best substrate for acclimatization was Plantmax® mixed with carbonized rice peels and 1.68 g per cell of the Osmocote® fertilizer.
