Eighteen Arabica coffee lines derivative from the genotype C1195-5-6-2, and the cultivars IAPAR 59 and IPR 99 were evaluated for resistance to bacterial halo blight in two trials carried out in field conditions, in Londrina, PR, Brazil. The cultivars Mundo Novo IAC 376-4 and Catuaí Vermelho IAC 81 were included as susceptible controls. Ten lines and the cultivar IAPAR 59 showed resistance to bacterial halo blight. The cultivar IPR 99 presented intermediate reaction, and the controls were very susceptible. This is the first study to show that lines derivative from the genotype C1195-5-6-2, which has C. racemosa genes, could be a source of resistance to bacterial halo blight in coffee breeding programs.Item Assisted selection using molecular markers linked to rust resistance SH3 gene in Coffea arabica(Crop Breeding and Applied Biotechnology, 2023-10-25) Silva, Angelita Garbossi; Ariyoshi, Caroline; Shigueoka, Luciana Harumi; Pereira, Luiz Filipe Protasio; Sera, Gustavo HiroshiThe aim of this work was to validate markers linked to the SH3 gene of coffee leaf rust (CLR) resistance and use them in assisted selection. Initially, we validated the markers in genotypes already known to carry SH3. Next, we performed phenotype and genotype evaluation for resistance to CLR in coffee plants growing under field conditions. We used Arabica coffee progenies derived from BA-10, which carries the SH3 gene due to introgression of C. liberica. Three SCAR markers (SP-M16-SH3, BA-48-21O-f, and BA-124-12K-f) and one SSR marker (Sat244) linked to SH3 gene were used to amplify the coffee plants’ DNA. Our assessments of markers validation in resistant genotypes, SP-M16-SH3 and BA-124-12K-f, were efficient to identify the SH3 gene. These two markers were used to evaluate the progenies derived from BA-10 and were significantly linked to the phenotype evaluations. The SP-M16-SH3 marker was more efficient, with the advantage of being codominant.Item IPR 106: new Arabica coffee cultivar, resistant to some Meloidogyne paranaensis and M. incognita nematode populations of Paraná(Crop Breeding and Applied Biotechnology, 2020) Sera, Gustavo Hiroshi; Machado, Andressa Cristina Zamboni; Ito, Dhalton Shiguer; Shigueoka, Luciana Harumi; Silva, Santino Aleandro da; Sera, TumoruCultivar IPR 106 resulted from a spontaneous hybridization between “Icatu IAC 925” and an unknow dwarf plant. It is a dwarf cultivar with high rusticity, late ripening cycle, large grains, excellent cup quality and resistance to some populations of the nematodes Meloidogyne paranaensis and M. incognita found in the state of Paraná.Item Associação de Pseudomonas syringae pv. garcae com algumas características agronômicas em cafeeiros F2 segregantes para o gene erecta(Editora UFLA, 2004-09) Sera, Gustavo Hiroshi; Sera, Tumoru; Altéia, Marcos Zorzenom; Androcioli Filho, Armando; Azevedo, José Alves de; Petek, Marcos Rafael; Ito, Dhalton ShiguerA bacteriose causada pela Pseudomonas syringae pv. garcae provoca grandes prejuízos em algumas regiões da cafei- cultura brasileira, como Paraná, São Paulo e Minas Gerais, principalmente em lavouras novas, podadas e expostas ao vento. Com este trabalho, objetivou-se estudar nas plantas F 2 do cruzamento entre os genótipos IAPAR-59 e Catuaí Erecta a associação entre as características vigor vegetativo e ângulos de inserção das ramificações plagiotrópicas sobre a intensidade de ocorrência da bacteriose. Avaliaram-se, em outubro de 2001, 316 plantas F 2 plantadas no IAPAR de Londrina em outubro de 1998. A escala de notas de avaliação da incidência de bacteriose adotada foi de 1 a 5, sendo 1 = plantas sem lesão e 5 = plantas com muitas lesões. As notas de vigor vegetativo adotadas foram de 1 a 5, sendo 1 para folhas de coloração verde-claras e 5 para folhas verde- escuras. Para a inserção dos ramos plagiotrópicos sobre os ortotrópicos, as notas variaram de 1 a 3, sendo 1 = normal (45 a 70o), 2 = semi-ereta (25 a 40o) e 3 = ereta (5 a 20o). Estimou-se o coeficiente de correlação de Pearson para verificar a associação entre as características vigor vegetativo e do ângulo de inserção das ramificações plagiotrópicas com a intensidade de ocorrência da bacteriose nos cafeeiros. A correlação estimada entre a intensidade da bacteriose com o ângulo de inserção dos ramos plagiotrópicos foi de r = + 0,2087**. Não houve correlação significativa entre a bacteriose e o vigor vegetativo. Assim, plantas com ramificação plagiotrópica ereta são predispostas a uma maior incidência de bacteriose.Item Coffee berry borer resistance in coffee genotypes(Instituto de Tecnologia do Paraná - Tecpar, 2010-03) Sera, Gustavo Hiroshi; Sera, Tumoru; Ito, Dhalton Shiguer; Ribeiro Filho, Claudionor; Villacorta, Amador; Kanayama, Fabio Seidi; Alegre, Clayton Ribeiro; Grossi, Leandro DelThe aim of this study was to evaluate the coffee germplasm of the Paraná Agronomic Institute (IAPAR) for resistance to the coffee-berry-borer. Preliminary field evaluation was performed in August 2004 and the fruits of less damaged genotypes in the field were evaluated under controlled condition with obligated and free choice experiments established in a randomized complete design with three replications. The genotypes were evaluated fifteen days after infestation with one borer per fruit in Petri dishes. The data were analyzed by the Scott-Knott means test at 1 % and by the χ 2 test. Statistical analysis indicated that Coffea kapakata, Psilanthus bengalensis, C. eugenioides and genotypes with C. eugenioides genes were resistant. These genotypes presented low frequency of bored grains. C. eugenioides and C. kapakata could present resistance at epicarp level but not in the grain. Twenty one CIFC’s coffee trees with several resistance genes SH were evaluated in field conditions. All the evaluated coffees carrying SH3 gene presented resistance to the rust. It was possible that rust races with the virulence gene v3 in the Paraná State didn’t exist. The SH3 gene in combination with genes SH5, SH6, SH7, SH8, SH9 and SH? would be very important to obtain cultivars with more durable resistance to the rust.Item Selection for durable resistance to leaf rust using test-crosses on IAPAR-59 and Tupi IAC 1669-33 cultivars of Coffea arabica(Instituto de Tecnologia do Paraná - Tecpar, 2007-07) Sera, Gustavo Hiroshi; Sera, Tumoru; Ito, Dhalton Shiguer; Azevedo, José Alves de; Mata, João Siqueira da; Doi, Deisy Saori; Ribeiro Filho, ClaudionorThe aim of this study was to identify plants of the IAPAR-59 and Tupi IAC 1669-33 coffee cultivars with less defeated resistance genes by the rust races present at IAPAR (Londrina, Paraná State, Brazil) using test-crosses. Eighteen test-crosses derived from hybridizations between ‘IAPAR-59’ or ‘Tupi IAC 1669-33’ with susceptible coffee to the rust disease were evaluated. Six hybrids were used as susceptible standards originated from hybridizations between two susceptible coffee plants. Many parental plants of the ‘IAPAR-59’ and ‘Tupi IAC 1669- 33’ presented more defeated resistance genes against rust races present at IAPAR than others of these cultivars or the genes were in heterozygous, because of segregant susceptible plants observed in some test-crosses. The test- crosses were very efficient to identify plants with less defeated resistance genes to the H. vastatrix. Coffee plants considered resistants would must be made test-crosses to verify which plants presented less and/or more defeated genes in homozygous.Item Rust resistance in arabic coffee cultivars in northern Paraná(Instituto de Tecnologia do Paraná - Tecpar, 2013-01) Grossi, Leandro Del; Sera, Tumoru; Sera, Gustavo Hiroshi; Fonseca, Inês Cristina de Batista; Ito, Dhalton Shiguer; Shigueoka, Luciana Harumi; Andreazi, ElderThe objective of the study was to evaluate the resistance to rust in coffee cultivars developed by research institutes of Brazil in Paraná state. Resistance to the local leaf rust races was assessed in high disease intensity field conditions at Londrina and Congonhinhas in 2009 and 2010. The cultivars were developed by the EPAMIG/UFV, IAPAR, IAC and MAPA/Procafé. The resistant standard ‘IAPAR 59' and the susceptible standards Catuaí Vermelho IAC 144' and 'Bourbon Amarelo' were used. A randomized block design with three replications and plots with 10 plants was used. A scale from 1 to 5 based on the rust intensity was used to evaluate the resistance. The Catiguá MG 1, Catiguá MG 2, IAPAR 59, IPR 98, IPR 104, Palma II, Paraíso H-419-10-6-2-5-1, Paraíso H-419- 10-6-2-10-1, Paraíso H-419-10-6-2-12-1, Pau Brasil MG 1 and Sacramento MG 1 cultivars presented complete resistance to rust at Londrina and Congonhinhas. The cultivars derived from the Catucaí germplasm were susceptible or showed different levels of partial resistance. Partial resistance to the rust was observed in several coffees derived from “Hibrido de Timor”. 'Acauã' and 'Obatã IAC 1669-20' presented complete resistance at Londrina, but at Congonhinhas, they were partially resistant, indicating that different rust races have occurred at these two locations.Item Partial resistance to fruit necrosis associated to Colletotrichum spp. among arabic coffee genotypes(Instituto de Tecnologia do Paraná - Tecpar, 2007-05) Sera, Gustavo Hiroshi; Sera, Tumoru; Ito, Dhalton Shiguer; Azevedo, José Alves de; Ribeiro Filho, Claudionor; Mata, João Siqueira daThe aim of this study was to evaluate the resistance to fruit necrosis associated to Colletotrichum spp. in Coffea arabica L.. Twenty-two arabic coffee genotypes were performed in a randomized block design, with three replications and ten plants per plot. They were evaluated for the variables fruit necrosis, luminosity index inside of the plant canopy and fruits per productive nodes of the plants. There was genetic variability for the resistance to fruit necrosis. Different partial resistance levels, varying from highly susceptible (score = 3.89) to moderately resistant (score = 1.77) were observed.
