    Microsatellite DNA fingerprinting of Coffea sp. germplasm conserved in Costa Rica through singleplex and multiplex PCR

    A large collection of coffee genetic resources is conserved in Costa Rica. In this study, microsatellite DNA fingerprinting of coffee through singleplex and multiplex PCR approaches coupled with capillary electrophoresis are described. To validate both methods, germplasm of Coffea spp. (Arabica and non-Arabica) and intraspecific F1 hybrids were analyzed using fourteen microsatellite markers. It was observed that through both PCR methods the fingerprinting profile of a subset of samples was identical. The genetic analyses revealed that non-Arabica coffee displayed greater genetic variation than Arabica coffee did. In addition, microsatellite analyses allowed the separation of C. arabica from other species using the principal coordinate analysis (PCoA) approach. The neighbor-joining tree clustering analysis revealed either a grouping of wild genotypes separated from cultivars of C. arabica, or a relation of intraspecific F1 hybrids with parental lines. The utility of our methodology for the characterization of F1 hybrids not previously analyzed through SSR (Simple Sequence Repeats) fingerprinting is demonstrated.

    Nutritional efficiency in phosphorus of arabica coffee genotypes

    Nutritional efficiency is a term used to characterize plants in their capacity to uptake and use nutrients, being related to the efficiency of uptake, translocation, and utilization of nutrients. Different coffee genotypes are expected to exhibit variability in their nutritional efficiency. This work aimed to evaluate the phosphorus nutritional efficiency in arabica coffee genotypes. The experiment was carried out in a greenhouse of the Empresa de Pesquisa Agropecuária de Minas Gerais (EPAMIG), located in the municipality of Lavras – Minas Gerais, Brazil. The experimental design was a randomized complete block design, in a 10x2 factorial scheme (10 arabica coffee genotypes and two dosages of phosphate fertilization), with four replicates. Each experimental plot consisted of one pot with ten liters of soil, with one plant. The cultivars Catiguá MG2, MGS Ametista and Sarchimor MG 8840 are neither efficient nor responsive to phosphate fertilization. Progeny H 6-47-10 pl. 3 and the cultivar Catuaí Vermelho IAC 144 are not efficient but are responsive to phosphate fertilization. The cultivars Paraíso MG H 419-1, Topázio MG 1190 and MGS Paraíso 2 are efficient but are not responsive to phosphate fertilization. The cultivars Bourbon Amarelo IAC J10 and MGS Aranãs are efficient and responsive to phosphate fertilization.

    Associação de Pseudomonas syringae pv. garcae com algumas características agronômicas em cafeeiros F2 segregantes para o gene erecta

    A bacteriose causada pela Pseudomonas syringae pv. garcae provoca grandes prejuízos em algumas regiões da cafei- cultura brasileira, como Paraná, São Paulo e Minas Gerais, principalmente em lavouras novas, podadas e expostas ao vento. Com este trabalho, objetivou-se estudar nas plantas F 2 do cruzamento entre os genótipos IAPAR-59 e Catuaí Erecta a associação entre as características vigor vegetativo e ângulos de inserção das ramificações plagiotrópicas sobre a intensidade de ocorrência da bacteriose. Avaliaram-se, em outubro de 2001, 316 plantas F 2 plantadas no IAPAR de Londrina em outubro de 1998. A escala de notas de avaliação da incidência de bacteriose adotada foi de 1 a 5, sendo 1 = plantas sem lesão e 5 = plantas com muitas lesões. As notas de vigor vegetativo adotadas foram de 1 a 5, sendo 1 para folhas de coloração verde-claras e 5 para folhas verde- escuras. Para a inserção dos ramos plagiotrópicos sobre os ortotrópicos, as notas variaram de 1 a 3, sendo 1 = normal (45 a 70o), 2 = semi-ereta (25 a 40o) e 3 = ereta (5 a 20o). Estimou-se o coeficiente de correlação de Pearson para verificar a associação entre as características vigor vegetativo e do ângulo de inserção das ramificações plagiotrópicas com a intensidade de ocorrência da bacteriose nos cafeeiros. A correlação estimada entre a intensidade da bacteriose com o ângulo de inserção dos ramos plagiotrópicos foi de r = + 0,2087**. Não houve correlação significativa entre a bacteriose e o vigor vegetativo. Assim, plantas com ramificação plagiotrópica ereta são predispostas a uma maior incidência de bacteriose.

    Coffee berry borer resistance in coffee genotypes

    The aim of this study was to evaluate the coffee germplasm of the Paraná Agronomic Institute (IAPAR) for resistance to the coffee-berry-borer. Preliminary field evaluation was performed in August 2004 and the fruits of less damaged genotypes in the field were evaluated under controlled condition with obligated and free choice experiments established in a randomized complete design with three replications. The genotypes were evaluated fifteen days after infestation with one borer per fruit in Petri dishes. The data were analyzed by the Scott-Knott means test at 1 % and by the χ 2 test. Statistical analysis indicated that Coffea kapakata, Psilanthus bengalensis, C. eugenioides and genotypes with C. eugenioides genes were resistant. These genotypes presented low frequency of bored grains. C. eugenioides and C. kapakata could present resistance at epicarp level but not in the grain. P. bengalensis could present resistance also in the grains.

    Resistance to leaf rust in coffee carrying SH3 gene and others SH genes

    The aim of this work was to evaluate the resistance to rust in coffee carrying SH3 gene and other SH genes. Twenty one CIFC’s coffee trees with several resistance genes SH were evaluated in field conditions. All the evaluated coffees carrying SH3 gene presented resistance to the rust. It was possible that rust races with the virulence gene v3 in the Paraná State didn’t exist. The SH3 gene in combination with genes SH5, SH6, SH7, SH8, SH9 and SH? would be very important to obtain cultivars with more durable resistance to the rust.

    Partial resistance to fruit necrosis associated to Colletotrichum spp. among arabic coffee genotypes

    The aim of this study was to evaluate the resistance to fruit necrosis associated to Colletotrichum spp. in Coffea arabica L.. Twenty-two arabic coffee genotypes were performed in a randomized block design, with three replications and ten plants per plot. They were evaluated for the variables fruit necrosis, luminosity index inside of the plant canopy and fruits per productive nodes of the plants. There was genetic variability for the resistance to fruit necrosis. Different partial resistance levels, varying from highly susceptible (score = 3.89) to moderately resistant (score = 1.77) were observed. The genotypes that presented moderate resistance were the cultivars IPR- 100, IPR-103, IPR-105 and IPR-108 and the coffee selection IAPAR-00023.

    Selection of Arabic coffee progenies with rust resistance

    The aim of this study was to select high-yielding coffee progenies with resistance to coffee leaf rust for the State of Paraná (Brazil). Field experiments in a randomized block design were carried out in Itaguajé and Congonhinhas. Yield, vegetative vigor and rust resistance of nine progenies of Arabic coffee and three check cultivars were evaluated. Many genotypes derived from “Sarchi- mor” and “Catucaí” were susceptible. Three coffee genotypes of Sarchimor germplasm and the F 6 generation of genotype “Catuaí x (Catuaí x BA-10 coffee)” were selected to advance generations and are promising to become new cultivars for being higher-yielding than ‘IAPAR 59’ and ‘Tupi IAC 1669-33’ and having a large number of plants with complete rust resistance.
